Protocols of antioxidant enzymes and chlorophyll fluorescence

Dear colleagues,
I found this website very useful after www.plantstress.com(external link)
Since I am also running a web based group Plant Stress Physiology titled as PlantStress

I would like to highly acclaimed its usefulness. However, it is requested that most of plant physiologists and molecular biologists are focusing oxidative stress responses in plants under salt stress (Munns and Tester, 2008), water stress, heavy metal stress, flooding stress, abiotic stress (Apel and Hirt, 2004), it is necessary to upload lab protocols for how to extract antioxidant enzymes, SOD, CAT, POD, GSH etc and how calculations were made. I have also done this job with help of my colleagues in another institution.

Similarly, Chlorophyll fluorescence is another important tool for screening for stress tolerance in breeding program as well as to find out underlying mechanisms. Since people are using different nomenclature OJIP curves etc using different instruments it is also necessary to put detail protocols for this

Dear Habib,

You might find the protocol on Imaging PAM Chlorophyll Fluorometer(external link) useful. This is the only protocol on chlorophyll fluorescence available at this stage, so any further contributions will be very welcome!

Dear colleagues,

I am in search of protocol for chlorophyll fluorescence using mini PAM in field or glasshouse. No doubt thermal imaging is a very good technique, but its too costly.

2ndly I am in search of lab protocols for antioxidant enzymes. Although I have a number of protocols published in research articles, I might doing some error/wrong step due to which results are not satisfactory/reproducible.

Dear Dr Habib

That is a very good idea to include protocols on extracting antioxidant enzymes. I will contact the editor in charge of Tissue Chemistry and we will invite some protocols.

I would like to add that in the Munns and Tester (2008) review, we cautioned about the physiological interpretation of changes in antioxidants, which can be secondary responses to the stress. These measurements need to be accompanied by measurements of growth rate and of photosynthesis. Otherwise we do not know whether the changes in antioxidants indicate tolerance to the stress, or sensitivity to it.
